This antibody detects membrane-bound latent TGF-b1 by staining of the latency associated peptide (LAP). I use only 5ul instead of the recommended 20ul but it works beautifully. I normally stain for 20min at 4'C. The cells can be fixed afterwards if necessary but I don't recommend permeabilization.

LAP is hardly expressed on normal PBMCs but we found it on CD4+ T cells of patients with Multiple Sclerosis (see image). We normally use both FMO and isotype control to verify the data.

Great antibody, but I recommend using FMO and/ or isotype control in parallel.
