Our laboratory work with human cancer cells, performing a lot of confocal analysis for golgi localization of various glycosyltrasferase enzymes. It is very essential to use a very good golgi marker in order to establish precise localization of glycosyltransferases. The abcam "anti-giantin" antibody has always given us good, consistent and reproducible staining though the cells express least amount of protein.

Dilution-1:200 in blocking agent for 60 mins at room temperature

5% Normal donkey serum prepared in PBST. (137mM NaCl, 2.7mM KCl, 8mM Na2HPO4, 1.46mM KH2PO4, 0.05% Tween 20)

Secondary Incubation

Donkey anti-rabbit (JIR:Cat#711-585-152), Dil-1:500 in blocking buffer, Time:30 min at room temp

Tertiary Incubation

None

Fluorescence and Confocal microscopy

Results Summary

We have run this experiment several times with this anti-giantin antibody. Each time we obtained high quality results which were used to publish the results. We have also used this antibody for tissue staining as well as western blotting with very good signal.

Additional Notes

Need lower concentration for western blotting (1:1000 Dilution)
