TAE Buffers

TAE buffer, also known as Tris-acetate-EDTA, is a running buffer routinely used for the electrophoresis of nucleic acids in a gel matrix such as agarose or polyacrylamide. It comprises of Tris base, EDTA, and acetic acid, which gives it a suitable ionic strength. This is vital for conducting electricity and maintaining the pH. The buffer ensures that the overall net charge of the nucleic acid is uninterrupted, thus facilitating their movement across the gel matrix. TAE buffer is highly recommended when working with RNA and DNA fragments larger than 1500 bp or for working with genomic DNA during cloning. TAE buffer can be used as both a running and gel preparation buffer. It is ideal due to its relatively easy and quick preparation and also because double-stranded DNA has a tendency to run faster in TAE.
