Quantifying Immunotherapy Impact with Multiple Analysis Techniques

Cancer immunotherapies are increasingly being explored as a method to harness the immune system to attack cancer cells. For such studies, the ability to correlate T cell biomarker expression and secretion with target cell killing is critical. Moving this research from the bench to the clinic is critically important, but reliable tools are needed to translate in vitro protocols and results to in vivo ones. In this application note, a promising new class of therapeutics called bispecific T-cell engagers (biTEs) is evaluated. The capability of CD19-BiTEs to enhance the cytotoxic effects of T lymphocytes on a B-Cancer cell line, Daudi cells, is studied using three distinct assays. Target cell death was monitored using an RTCA impedance assay and a flow cytometry-based cytolysis assay; while secretion of cytokines and cytolytic proteins were quantified in a bead-based multiplex flow cytometry assay to monitor the T cell response.
