The MTT cell viability assay is designed to evaluate the metabolic activity and viability of cells. The assay involves the conversion of the yellow tetrazolium dye MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) to an insoluble purple formazan. The enzymes responsible for the reaction are NAD(P)H-dependent cellular oxidoreductases, which are only active in viable cells. The absorbance of the colored solution can be quantified using a spectrophotometer and will be directly proportional to the concentration of formazan.
