Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of B cells and antibody-secreting cells (ASC) in health and disease, in this experiment we are looking for markers of PC, but we stain CD4+ T cells to be sure the antibody works.

First, we gate lymphocytes and single cells using side scatter, forward scatter and exclude dead cells using PI (PI- cells).Then, we gate on CD4+ T cells. Few of the CD4 T cells express CD365. We use a FMO control to make sure the staining is specific.
