Currently we are making an assessment of the microglial response in several areas of the basal ganglia following the induction of a pharmacological model of Parkinson´s Disease.We employed this antibody, to perform both characterization of the cell densities as well as fluorescence intensity and morphometrical analysis.
Immunofluorescence
Brain Tissue, PFA fixed, Vibratome processed
1:50 dilution with 12 hours incubation at 4ºC under agitation.
10%NGS in PBS-Triton 0.5% for 1,5 hours
Alexa Fluor 488, 1:1000 for 2 hours

Confocal Microscopy
We were able to evidence, intense microglial morphological change in the ipsilateral side (drug-treated) animals.In addition, cell number increased significantly and focally at substantia nigra pars compacta and reticulata.Our treatment paradigms reduced such microglial responses.
