Annexin V staining is the most reliable tool to detect cells undergoing apoptosis. Biolegend provides Annexin V protein with various fluorescent tags so that it is very useful to analyze cell apoptosis together with other dyes or antibodies. FITC annexin V is compatible with 7-AAD, a cell impermeable nucleic acid dye that is frequently used together in flow cytometry and FITC conjugation is the least inexpensive option.

For Annexing V staining, home-made buffer (10-20mM HEPES saline with 1-3mM CaCl2) works as well as any commercially available buffer

Results Summary

Attached assays are showing the progression of apoptosis in mouse neutrophils. Bone marrow-derived neutrophils (100,000 cells) were incubated with FasL for various periods of time. The cells were then stained with 2.5 ul of the reagent in 100 ul buffer for 15 min at 4 degree.
