I use this cell line to establish small-airway like air-liquid interface cultures. Primary human leukocytes are then migrated through the epithelium as a model for recruitment to the lung.

Tips

Grows quickly after several passages but can be slow to revive from freezing. Consider freezing 2x10^6 cells per vial and plating in 20% FBS until, then reducing to 10% once desired doubling time is reached.

Results Summary

This cell line is grown in standard culture methods and then grown at air-liquid interface on Alvetex filters similar to a transwell. Human neutrophils are then isolated from blood and migrated through the epithelium using desired chemoattractants/stimuli to model various conditions of recruitment into the lung. This cell line is reliable through at least 25 passages and can tolerate various disturbances such as the occasional forgotten media change or incubator running out of CO2 for a few hours.
