Product Name:

Brilliant Violet 650 anti-human CD63

Catalog Number:

353026

Clone Number:

H5C6

I used this antibody to stain for CD63, a marker of primary granule exocytosis, on neutrophils from blood and sputum of an adult CF patient

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Blood, sputum

Primary Incubation

2.5 uL of antibody added to 2x10^5 cells in 50 uL of PBS+EDTA

Blocking Agent

Fc receptor blocking before antibody stain

Secondary Incubation

none

Tertiary Incubation

none

Detection

Flow cytometry; V670/30 filter on BD Symphony

Results Summary

I used this antibody to compare degranulation by blood and airway neutrophils. Blood neutrophils are mostly negative for CD63, indicating minimal exocytosis of primary granules. Airway neutrophils from sputum demonstrate 3-4 log increase in expression of CD63, indicating significant primary granule exocytosis.
