We regularly use this kit to isolate genomic DNA of bacterial transposon libraries. Because there are many unique mutants throughout the library, we need a high genomic DNA yield of each sample to have good representation of the mutant ratios present in the library.

This kit works well because it gives a high yield which allows us to isolate gDNA from a large number of bacteria within the library.Genomic DNA collected from this kit shows minimal denaturation, leaving the majority of the gDNA as a large product at the top of the gel.

The Bottom Line

This is a useful kit for genomic DNA isolation of high complexity samples for high throughput sequencing. It works for both Gram positive and Gram negative bacteria and provides a high yield for both, provided that you include a bead beating step for the Gram positive.
