I use this kit to determine knockdown of TGFb-1 from cell supernatent of my in-vitro experiments. I have tested this kit for both mouse and human cell lines and it works perfectly.

Results Summary

Cell supernatent from both human (HepG2) and Mouse (AML-12 and RAW264.7) cell lines were tested for TGFb-1 expression. The aim of the experiment was to determine the baseline expression of TGFb-1 secreted by the cells. TGFb-1 knockdown was assessed 96 hours post transfection with ELISA. Also, to reduce the background from FCS, the cells were adapted to serum-free conditions and appropriate media-only controls were run during the experiment.

The kit is quite straight forward to use, the protocol provided is not at all complicated. Although, the ELISA takes about 3 days to finish as two overnight steps are involved. The kit is quite sensitive, I could measure as low as 10pg/ml concentrations of TGFb-1 from the cell supernatent which is quite useful. For the maximum sensitivity it is recommended to let the antigen bind overnight at 4 degree C than at room temperature for 2 hours. I have tried both and overnight works the best. Also this kit is quite inexpensive as compared to other coated ELISA kits. I did get quite reproducible results and hence the kit is very reliable for a series of experiment.
