The aim of the experiment was to measure GLP-1 levels secreted by STC-1 cell line (enteroendocrine cells) in response to protein hydrolysates. 1.2 × 106cells were seeded into 6 well plates in DMEM media for 24 hrs at 37°C in 5% CO2 , then washed 1-2 times with HEPES buffer. After 1 h, HEPES buffer is aspirated and 1 ml casein and whey protein hydrolysates (100μg/ml) solution were added to wells. Cells were then incubated for 4 hrs at 37°C, 5% CO2. The supernatant was collected carefully and GLP-1 was measured according to manufacturer’s protocol.

GLP-1 was measured in supernatant. The concentration of GLP-1 is calculated by the standard curve and % GLP-1 secretion was calculated by the following formula: % GLP-1 secretion: [(OD Test sample –OD Blank)/(OD Control-OD Blank)]*100Results obtained were consistent and highly reproducible.

Easy to perform the experiment
The procedure takes approximately 1.5 h to complete and multiple washing steps is also time consuming.
GLP-1 Elisa kit produce good results with less sample processing.
