Company:

New England Biolabs

Product Name:

Lambda Protein Phosphatase

Catalog Number:

P0753S

Good product with strong enzymatic action.

Experimental Design and Results Summary

Application

Western blotting

Starting Material

Mouse oocytes' lysate

Tips

Protein extracts from 200 oocytes or 200 eggs were incubated with 2 Units/μL of Lambda Protein Phosphatase (New England Biolabs, #P0753S) in 1X NE Buffer (50 mM HEPES, 100 mM NaCl, 2 mM DTT, 0.01% Brij 35, pH 7.5 at 25°C) supplemented with 1 mM MnCl2 at 30ºC for 2 hours. The protein extracts (21 micrograms) were then used for SDS-PAGE and Western blotting by following standard procedures.

Results Summary

This enzyme is used to detect phosphorylation status of proteins that can be phosphorylated at serine/ threonine/ tyrosine residues, like CDC25B.

Summary

The Good

Detects if a protein is phosphorylated at Ser/ Thr/ Tyr residues.

The Bad

Phosphorylation of the protein at other residues may not be detected by the product.

The Bottom Line

Great reagent for detection of protein phosphorylation, quick and easy to use.
