Good quality genomic DNA from Escherichia coli was isolated using Wizard Genomic DNA purification kit. With 1 ml of overnight grown culture, the yield was 15 microgram in 100 microliter elution with nuclease free molecular biology grade water. The quality of the eluted DNA was checked by Nanodrop Spectrophotometer and found as 260:280 ratio of 1.85 and 260:230 ratio of 2.10.

Starting Material

Escherichia coli grown in Luria Bertoni broth was used (1ml per prep)

Tips

Make sure to spin and pellet the bacterial cells and remove completely any media broth left.

Results Summary

E. coli culture grown overnight was used for the isolation of genomic DNA. 1 ml culture was used per prep and followed according to the kit's instructions. Also, included optional RNase digestion to remove RNA. Finally eluted with 100 microliter nuclease free water after isopropanol precipitation. Eluted genomic DNA was quantified by Nanodrop Spectrophotomer. The overall yield was found to be 15 microgram with 260:280 ratio of 1.85 and 260:230 ratio of 2.1.
