Our immunology lab studies CD8+ T cell differentiation. In order to work with isolated CD8+ T cells, we routinely perform cell separation of mouse splenocytes to obtain pure CD8+ T cells to perform downstream experiments. This isolation kit allows us to recover such cells with relative ease after optimization.

Tips

We ended up using a magnetic separation column to help with the purity and yield. Must use less as mentioned on their website since column allows less usage of antibody and beads!! FIlter mixture once more before applying to column to remove aggregates.

Results Summary

After optimizing based on starting sample, we used less antibody and beads then recommended for each isolation especially with transgenic mouse T cells. We use a LS column from Miltenyi but use this isolation kit which saves us a significant amount of money per year.

Summary

The Good

Very cheap compared to other vendors and results in good purity.

The Bad

Takes a bit longer (30 mins) compared to Miltenyi and requires use of LS column.

The Bottom Line

We are very happy in the end using this kit since it's very cheap per isolation. Yield and purity is very acceptable especially with transgenic mouse T cells.
