Sample: Human colon, 15 µm-thick section

Secondary Antibody: Alexa Fluor 488 Donkey anti-Rabbit IgG (H+L)
Catalog number: Life Technologies A-21206 (from Molecular Probes)

Block: 10% Normal donkey serum (NDS) in PBS
Incubation time: 60 min
Incubation temperature: 25°C

Primary Antibody Incubation Dilution used: 1/1000
Diluent composition: PBS+ NDS 1%
Incubation time: 24 hr
Incubation temperature: 4°C

Secondary Antibody Incubation
Dilution used: 1/500
Diluent composition: PBS + NDS 1%
Incubation time: 1 hr
Incubation temperature: 25°C

Detection Method: Confocal microscope (Leica TCS SP2) observation of tissue section mounted onto a slide.

Results Summary: I observed good staining in human colon epithelium cells and in smooth muscle cells (longitudinal and circular).
