I am researching immunotherapies for cancer in mice, and my general research aim is to determine the extent of immune cell infiltration as well as activation by staining cytokines. This viability dye is great for staining live cells that will then be fixed and permeabilised.

Tips

Make sure cells have been washed thoroughly with PBS to remove any proteins that will quench the labelling reaction

Results Summary

Live cells are stained violet but only on extracellular proteins. Dead cells are stained on extracellular and intracellular proteins, therefore dead cells show increased staining in the DAPI channel and can be excluded from analysis.

The Bottom Line

Follow the manufacturer's instructions to be able to stain live cells in flow cytometry for cells that need to be fixed and permeabilised to be able to stain intracellular proteins.
