Company:

Product Name:

Collagenase type IV

Catalog Number:

17104-019

Excellent dissociation reagent for human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs). We use it to regularly passage human pluripotent stem cells (hPSCs) as colonies, as it is a mild reagent that does not break them down into single cells.

Experimental Design and Results Summary

Application

Dissociation of human pluripotent stem cell colonies (hPSCs)

Starting Material

hPSCS, DMEM F12

Tips

For use with hPSCs when cultured on a feeder layer of mouse fibroblasts.

Results Summary

We use Collagenase type IV to preserve stem cell colony morphology in the range of 1-2 mg/mL, diluted in DMEM F12

Summary

The Good

Easy dissociation process, gentle to the cells.

The Bad

Need to weigh first, then filter sterilize the diluted in DMEM F12 solution before use.

The Bottom Line

Working well with all stem cell lines we have in the lab, when cultured on feeder mouse embryonic fibroblasts (MEFs). Remains active in solution for a few days, so its possible to make some extra and then use within the next 10 days. If in a hurry, just use at 2mg/mL to shorten incubation time.
