I could study the activity of Hedgehog pathway by monitoring the protein level of Gli1. The MEF cells were arrested at the G0 phase, and treated or not treated with the ShhN-containing medium overnight. The cells were then lysed, and processed for WB. This antibody detects a protein other than Gli1 at around the expected molecular weight!
