We transformed 1ul of ligation mix into one shot TOP10 Electrocompetent, kept on ice for 5 min and provide pulse by electroporator. Added LB medium and incubated for 1 hour at 37c. Plated 10 ul of LB medium on to LB+antibiotic plates and incubated O/N at 37c. Observed well-spaced single colonies on all the transfected plates.
