The time course effect of a growth factor treatment on CD36 protein levels was measured in human endothelial cells. In brief, the growth synchronized cells were exposed to a growth factor for the indicated period, and the whole-cell extract was prepared. After protein quantification, an equal amount of proteins was resolved in SDS-PAGE and western transferred. The transferred proteins in the nitrocellulose membrane were immunoprobed with CD36 antibody obtained from Santacruz Biotechnology Inc. The antibody selection was based on flyer information.

Experimental Design and Results Summary

Applications

Western Blot

Sample

The human whole cell lysate

Primary Incubation

Dilution (1:250) in 5% skimmed milk prepared in 1x TBST, overnight incubation at 4 degree Celsius

Blocking Agent

5% skimmed milk made in 1x TBST

Secondary Incubation

Goat anti-mouse IgG HRP Ab, dilution (1:8000) in 5% skimmed milk made in 1x TBST, 1hr at RT

Tertiary Incubation

None

Detection

Enhanced chemiluminescent detection in X-Ray films

Results Summary

A highly specific and bright protein band of its desired molecular weight was detected. However, no effect of the growth factor on CD36 protein levels was observed.
