To study the apoptosis pathway during toxicity of fungal toxin, we have checked the apoptotic nuclei in control and toxin treated human keratinocytes (HaCaT) using Hoechst 33342 dye. Hoechst is a cell-permeable dye which stains the nucleus of both live and dead cells by intercalating in A-T region of DNA. Human keratinocytes were incubated with 10µg/ml Hoechst 33342 dye for 10 min at 37ºC in dark, intact and fragmented cell nuclei was visualized in fluorescence microscope under UV filter.
