Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of antibody-secreting cells (ASC) and B cells in health and disease. Here we use anti-CD3 to exclude T cells from our analysis.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Peritoneal Cavity cells from a C57Bl/6

Primary Incubation

Surface staining with anti-CD19, anti-CD5 and anti-CD3 BV510 (1/100), 20 min at 4C

Blocking Agent

Fc block (2.4G2 clone)

Secondary Incubation

N/A

Detection

Flow cytometer with a violet laser

Results Summary

As it can be seen in the figure, not all T cells (CD5hi in red) are stained and the staining is very weak. At least it seems to be specific and no CD19+ cells are stained.
