Tobacco mosaic virus (TMV) particles are potent vaccine candidates. We sought to determine if TMV is taken up via endocytosis in antigen presenting cells, using fluorescent TMV and the vital endosomal dye Lysotracker Deep Red under fluorescence microscopy.

D2/SC1 murine dendritic cells
We use LysoTracker at 1:100 dilutions. Cells were incubated with dye for 15-30 mins in 200 uL culture media, washed 1x, and replated in 250-500 uL of DPBS.
Endosomal compartments fluoresce red while FITC-labelled TMV were visualized in green (FITC) channels. The data show that TMV clearly enters endosomes.

Excess has to be washed out of culture in order to avoid high background; cannot be used on fixed cells.
