Company:

Thermo Fisher Scientific

Product Name:

IL-13 Monoclonal Antibody (85BRD), FITC, eBioscience™

Catalog Number:

11-7136-42

Clone Number:

85BRD

I tried a lot of IL-13 antibodies, and even though it's low, this is the only one I could get a consistent reasonable signal above background. The assay is finnicky and only worked using PMA and Ionomycin for 24 hours with a combination of BFA and Monensin. In addition, I had to use the eBioscience FoxP3 fix/perm buffer.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Frozen human PBMCs

Primary Incubation

surface stain 30 min room temp (for CD markers)

Blocking Agent

PBS with 1% FCS

Secondary Incubation

60 min on ice

Tertiary Incubation

Detection

Flow Cytometry

Results Summary

I've attached my titration and my time course for optimizing this protocol. The negative population is pretty bright but it's tight so you can still detect a positive population.

The Bottom Line

IL-13 is hard to detect and this is as good as I could find.
