I used this kit to quantify MCP-1 protein before and after inflammation in vitro in RAW 264.7 cells and 3T3 L1 cell lines. The levels of MCP-1 are expected to increase following the treatment with a pro-inflammatory reagent.
To quantify CCL2 in cell culture media
Centrifuged cell culture media supernatant
Perform washing steps at-least 6-7 times to get a clearer background with 60 sec. wait period in between each washing
Had to perform the analysis twice initially as the background wasn't clear. Overall, good reproducible estimation.
