In the present study, rat kidney tissue was harvested, micro sections were made and immunostained for pErk 1/2 proteins. Same tissue sections were stained for nuclear stain Hoechst 33342 trihydrochloride trihydrate and images were captured in a fluorescent microscope.
Fluorescent staining of cell nucleus in rat tissue sections
Rat pancreatic tissue sections, PBS, glass slide, OCT compound, primary and secondary Ab, fluorescent microscope etc.
Rats of 8–10 weeks age were sacrificed, the pancreas was harvested, fixed in 4% paraformaldehyde (w/v) solution, embedded in OCT compound, and tissue sections were made. Sections were immunofluorescently stained with specific antibodies. After secondary antibody treatment, same sections were treated with Hoechst 33342 trihydrochloride trihydrate reagent (dilution 1:1000 in PBS) for 4 minutes. After treatment sections were washed 3 times with PBS for 5 min each. After washing, sections were mounted on slides and observed in a fluorescence microscope.
