Here we were investigating if a novel cytosolic repressor of the Wnt pathway was causing degradation of Dvl2 or Dvl3. Samples with knockdown or overexpression of this repressor were stimulated with Wnt3a, and then Dvl3 was probed. The results were negative. Antibody staining by traditional western showed expected double band in the correct position; however, the signal was very weak.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Human breast cancer MDA-MB-231

Primary Incubation

1:500–1:1000 in 5% milk

Blocking Agent

5% milk in IBS

Secondary Incubation

Licor IRDye800-anti-mouse

Tertiary Incubation

None

Detection

Licor imaging

Results Summary

Results showed expected doublet band at the correct size with no non-specific bands. Signal was very weak and prevented robust quantification. Traditional HRP detection was not tried.
