The WNT pathway is essential for embryonic development and adult tissue homeostasis. Deregulation of this pathway results in cancers of the stomach, intestine, breast, etc. I am interested in studying the molecular mechanism of oncogenic WNT signal transduction to identify potential targets for therapeutic intervention of colon cancer. The goal of this experiment is to determine good working antibodies for the purpose of blotting some of the core components of the WNT pathway. This antibody is excellent for western blotting and picks up a nice clean band corresponding to GSK3beta.
Western Blot
RKO (whole-cell extract)
1:2000 dilution in 1xTBS-T; overnight at 4°C for 16 hr
5% milk in TBS-T
1:10,000 dilution of donkey anti-mouse IgG+HRP; 1 hr at room temperature
