The WNT pathway is essential for embryonic development and adult tissue homeostasis. Deregulation of this pathway results in cancers of the stomach, intestine, breast, etc. I am interested in studying the molecular mechanism of oncogenic WNT signal transduction to identify potential targets for therapeutic intervention of colon cancer. The goal of this experiment is to determine good working antibodies for the purpose of blotting some of the core components of the WNT pathway. This antibody was clean and didn't give any non-specific bands.

Experimental Design and Results Summary

Applications

Western Blot

Sample

RKO (whole-cell extract)

Primary Incubation

1:1000 dilution in 1xTBS-T; overnight at 4°C for 16 hr

Blocking Agent

5% BSA in TBS-T

Secondary Incubation

1:10,000 dilution of donkey anti-rabbit IgG+HRP; 1 hr at room temperature

Tertiary Incubation

None

Detection

ECL

Results Summary

RKO cells were treated with WNT3A for 0–2 hr and one can see the nice gradual accumulation of active beta-catenin.
