We aimed to study the affect of knocking down some demethylases in mouse prostate cells and change in any methylation pattern. GAPDH was used as the positive control.

Protocol Overview

The product, siRNA was used with the transfection reagent provided by Dharmacon, according to manufacturer's protocol. Cells were incubated from 72-96 hrs prior to detection of knockdown with western blot.

Tips

Don't overdose the cells. it works wonderfully at concentration mentioned by the manufacturer

Results Summary

About 90-100% of protein knockdown was observed. Worked as a great positive control, did not kill the cells.
