Company:

Santa Cruz Biotechnology

Product Name:

SLUG (A-7)

Catalog Number:

sc-166476

SLUG, the protein product of the SNAI2 gene, is a member of the Snail family of zinc-finger transcription factors. It is involved in the regulation of cell migration during embryogenesis and was implicated in regulation of tumor cell invasion and migration. We analyzed expression of SLUG in human carcinoma cell lines exhibiting different metastatic potentials. The mouse monoclonal antibody A-7 from Santa Cruz was employed in western blotting assays to detect SLUG in whole cell lysates under reduced conditions. One of the two protein bands recognized by this antibody has an apparent mol. wt. of ~34 kDa, which is consistent with the expected mol. wt. of SLUG.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Whole lysates of human carcinoma cells

Primary Incubation

Antibody at a final concentration of 1 microgram/ml, ON incubation with gentle rocking in a cold room

Blocking Agent

5% milk in PBS-0.05% Tween 20

Secondary Incubation

Stabilized goat anti mouse IgG HRP conjugate, 1:5,000 in blocking buffer

Tertiary Incubation

Detection

West Pico

Results Summary

In our hands, the mAb A-7 produces 2 major bands, one with an apparent mol. wt. ~34 kDa, consistent with the expected mol. mass of human SLUG. Unfortunately, the antibody also binds to a non-specific ~50 kDa protein with apparent mol. wt. of ~50 kDa.

Additional Notes

The image shows 2 major protein bands recognized by the SLUG antibody A-7. The lower band has an apparent mol. wt. of ~34 kDa, consistent with mol. wt. of human SLUG.

Summary

The Good

The antibody appears to be able to recognize human SLUG.

The Bad

The antibody appears to produce a non-specific proein band.

The Bottom Line

The mouse monoclonal antibody A-7 from Santa Cruz (sc-166476) could be used for detection of SLUG in whole lysates of human carcinoma cells in western blotting under reducing conditions. However, it should be taken into account this antibody also recognizes a non-specific protein band.
