Our lab works on engineering T cells to recognize and specifically kill HIV infected cells via lentiviral delivery of TCR genes. When TCR binds to the ligand on APC the cytoskeletal changes from the interaction and the binding lead to a series of downstream signals that result in increase of Ca2+ levels. Since classic killing assays do not always address minor differences between different TCRs, we perform assays measuring CA2+ levels to distinguish between TCRs.
