Integrins are cell surface transmembrane molecules that primarily mediate adhesion of cells to matrix proteins and induce cell signaling. Among integrins, beta 1 integrins play crucial roles in cell communications and migration of normal and cancer cells. Beta 1 integrins are also involved in stromal invasion and distant metastasis of cancer cells. We analyzed co-localization of beta 1 integrin with other cell surface receptors in human carcinoma cells. To achieve this goal, we verified the specificity and efficiency of MAB17781 to precipitate beta 1 integrins from human tumor cells .

MAB17781 nicely precipitated beta 1 integrin (~130 kDa) from control cells (Ctrl), but not from the cells in which beta1 integrin was knocked down (beta1 KD). As could be expected, less signal was obtained if control cells were trypsinized before their lysing. Immunoprecipitated beta 1 integrin was visualized after separation of IP protein complexes during SDS-PAGE under reducing conditions, and re-probing the blot with the MAB1965 from Millipore since MAB1965 can detect beta1 integrin under reducing conditions. Heavy (~50 kDa) and light (~25 kDa) chains of original MAB17781 used for IP are visualized by anti-mouse IgG conjugate.
