Our goal was to specifically label a subset of NKT cells in mice by flow cytometry and microscopy. We selected this marker based on a previous publication noting success. We tested different fluorophores, and found PE to be the best color due to the relatively low abundance of surface CD138.

Collagenase-digested mouse lung tissue
1:200 for 20 minutes on ice
24G2 for 10 minutes on ice
Fixed with 4% formaldehyde for 30 minutes

PE-conjugated CD138 stained our cells of interest specifically with no notable background. The separation was clear and robust.
