mTOR sub cellular localization is very critical for it's activation. mTORC1 activation takes place at the lysosomal surface of cells. During nutrient replete conditions, mTOR is readily observed at the lysosomal surface; however, during depleting conditions, mTOR is diffused throughout the cell. We were interested in observing this phenomenon in T cells during activation. We used this mounting solution for our microscopy studies to study mTOR localization. We used this solution since it didn't require overnight curing; therefore, allowing slides to be viewed soon after mounting with signaling protection.

Murine T cells
We used this mounting solution during the final step of the IF protocol. Depending on the surface area of the sample, we used an appropriate amount of solution. We dispersed the solution to ensure all areas were exposed then proceeded to place the cover slip on top before sealing.
Always be careful of bubbles from the small mounting solution vial.
Great consistent staining and preservation of signal. Avoids the additional DAPI step since it's within the mounting solution.

Easy to use mounting solution when time is critical since you can immediately view the slide after sealing. Relatively inexpensive if purchasing the larger pack.
