We study terminal differentiated B cells - plasma cells. These cells are also produced in structures called germinal centers. Therefore we stained for the presence of germinal center cells that are GL7+FAS+.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mesenteric lymph nodes of SPF and GF mice

Primary Incubation

GL7 FITC (1/200), anti-CD95 PE-Cy7, anti-CD19 incubate 15min at 4°C

Blocking Agent

Fc block with 2.4G2 30 µg/ml and incubate 15min at 4°C

Secondary Incubation

None

Tertiary Incubation

None

Detection

Flow cytometry

Results Summary

Mesenteric lymph nodes of GF mice present with a 10 fold reduction of Germinal Center B cells compare to SPF aged matched mice. Germinal center B cells are GL7+CD95+.
