Product Name:

P16-INK4A Antibody

Catalog Number:

10883-1-AP

We have identified a class of ATPase inhibitors that show promise for treatment of ovarian cancer. We are currently evaluating what mechanisms, if any, may lead to resistance to theses ATPase inhibitors. We are measuring changes in protein expression in an animal model of drug resistance. I chose this product because I have had success with Proteintech antibodies for numerous experiments.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Human tumor cell lysates

Primary Incubation

1:1000 dilution in 5% milk in TBS-T, overnight at 4 degrees C

Blocking Agent

5% milk in TBS-T

Secondary Incubation

1:10,000 in 5% milk in TBS-T, 30 minutes at RT

Tertiary Incubation

N/A

Detection

Chemiluminescence substrate for HRP-labeled secondary antibodies

Results Summary

In general, the antibody worked well and I could clearly identify a band at 16kDa. There are nonspecific bands on the blot - the product page does show this antibody may react with GAPDH. Overall this is an average antibody.

Summary

The Good

Good price, and Proteintech does sell trial sizes which is helpful.

The Bad

Multiple nonspecific bands observed

The Bottom Line

The antibody enabled me to generate the data I needed, but for publication I would probably try 1-2 additional antibodies to see if I can reduce nonspecific bands on the blot.
