I have constructed a expression plasmid called pACYC-cadA and transformed into to Invitrogen's One Shot Top10 chemically competent cells by heat shock method. After transformation, colonies with plasmid construct were selected on LB agar plates with Chloramphenicol at 10 microgram/ml. Transformation efficiency was very good (10^6 for microgram of plasmid construct being transformed). Cells with pACYC-cadA expressed my gene interest very well.

Starting Material

500 ng of the plasmid construct in a 50 microliter of One Shot Top10 chemically competent cells

Protocol Overview

Thaw One shot Top10 chemically competent cells on ice completely and then add plasmid construct (500 ng, ideally in less than 5 microliter volume). Mix the contents by flicking the bottom of the tube and then incubate on ice for 30 minutes. Heat shock the cells at 42 degree C for 45 seconds. Immediately place the heat shocked cells on ice for 5 minutes. Then recover these cells in 250 microliter of SOC medium at 37 degree C for 2 hrs. Plate appropriate dilutions of recovered transformation mix onto LB agar with chloramphenicol at 10 microgram/ml. Incubate plates at 37 degree C.

Tips

Make sure competent cells are thawed on ice completely before mixing it with plasmid DNA. Use pre-warmed SOC media at 37 degree C for recovery.

Results Summary

Transformation of plasmid construct using One Shot Top10 chemically competent cells went well with transformation efficiency of 10^6 cells/microgram of plasmid DNA
