This antibody was used in a study where induction of apoptosis in human tumor cells was followed up by detection of cleaved PARP-1 in western blots.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Human tumor cell lysates

Primary Incubation

Antibody at a final concentration of 1 microgram/ml, ON incubation with gentle rocking in a cold room

Blocking Agent

5% milk in PBS-0.05% Tween 20

Secondary Incubation

HRP-conjugated anti-mouse antibody at 1:5,000 in 5% milk in PBS-0.05% Tween 20

Tertiary Incubation

N/A

Detection

West Pico

Results Summary

This mouse monoclonal antibody, raised against PARP-1 C-terminus, detects with high efficiency both the full-length and cleaved PARP-1 in human tumor cell lysates.

Additional Notes

The image demonstrates protein bands corresponding to the full-length PARP-1 (~116 kDa) and cleaved PARP-1 (~89 kDa). Confirming high specificity of this antibody, the cleaved PARP-1 is detected only in the cells, which underwent apoptotic induction (right lane).
