Depleting a specific cell type in an experimental animal is extremely useful to investigate the functions specific for the cell. However, it is very challenging, so specific deletion is hardly attainable in most cases. Mouse neutrophil is an exception. Mouse neutrophils express a highly specific cell surface protein, Ly6G, and injection of anti-Ly6G antibody is a well-accepted method to delete neutrophils in a mouse.

Results Summary

Significant depletion of mouse neutrophils can be attained by one-time intraperitoneal injection of 300 - 500 ug of 1A8 antibody (anti-mouse Ly6G antibody) within 48 hours. The amount of antibody may be adjustable according to the mouse body weight and the efficiency may be even heightened by an additional secondary administration (such as 50-80 ug intratracheal administration for the study of airways). In the figure1A attached, neutrophils in spleen marked in dotted rectangle were barely detected in 1A8 injected mouse. In the figure 1B, neutrophils in influenza virus-infected lung and trachea were significantly depleted by 1A8 antibody.
