Our laboratory investigates ways to improve immune responses against HIV, including enhancement of the capacity of NK cells to kill HIV-infected cell. For these studies we set up in vitro ADCC assays with NK cells against infected target cells. As such, we need to purify NK cells and the CD56+ CD16+ selection kit isolates this population at a high purity via a two step process, first depleting all non-NK cells, and then enriching for the CD56 and CD16 positive cells.

We follow the kit protocol using the two step isolation, first depletion of non-NK cells, then enrichment of CD56+ CD16+ cells.

Tips

You can use this kit for the first step only, if it is important that NK cells are not activated or manipulated in any way.

Results Summary

The kit works very well. We get a very pure NK cell population and the CD56 CD16 enrichment step is very relevant for ADCC assays, given that CD16 (Fcgamma IIIa) is one of the Fc receptors activating NK cells to kill the target cells.
