Analysis of T cell subsets in humans requires the use of multicolor flow cytometry panels to identify distinct subpopulations by surface marker expression. Subsequent separation of CD3+ lymphocytes into CD4+ and CD8+ can then allow for specific characterization of immune responses. While CD3 is included in all of our multicolor panels, it is essential to save certain FACS channels for more difficult to detect antigens. We therefore selected this BUV395 conjugated CD3 antibody for the fluorophore's minimal spillover properties and to utilize the UV channel

Results Summary

Healthy human PBMCs were stained to analyze T cell subsets at steady state. We compared several different panels using the same antibodies conjugated to different sets of fluorophores to optimize detection of lowly expressed antigens. The CD3 BUV395 antibody enabled efficient detection of the T cell compartment within peripheral blood and exhibited good separation of signal following aldehyde fixation. Utilization of this antibody further allowed us to take advantage of the UV laser for our multicolor panels.
