Beta-actin has been broadly used in western blotting as a protein loading control. This post reviews the utility of a mouse mAb of beta-actin provided by Genscript. Overall this antibody is effective and much affordable compared to the antibodies from other producers.

Aim: Detection of beta-actin.Materials: 20ug of HEK-293T cell lysate, beta-actin antibody (0.5ug/ul stock)Methods: cell lysate was separated on a 10% SDS-PAGE gel and transferred onto a PVDF membrane. The membrane was blocked with 5% non-fat milk in TBST buffer at RT for 2 hours. Primary Ab incubation: 1:5000 diluted in TBST buffer, incubated at 4 degree overnight; Secondary Ab incubation: 1:10000 diluted in TBST buffer, incubated at RT for 1 hour. Result: beta-actin was clearly probed without non-specific band.
