I have been using BrightStar™ ECL Detection kit for my Western Blotting. I use this reagent kit especially when my blots have low concentrations of protein are when the antibody titer is very low. I get good bands of appropriate sizes without any background. The kit is cost effective and using this I have presented the data in many meetings. Soon the data obtained using this kit would be published. I have used this reagent kit since 2015 and they have worked well be is PVDF or nitrocellulose membrane. Alkali scientific Inc has good customer service.

Starting Material

Protein, nuclear and cytosolic lysates from mammalian prostate cancer cells

Protocol Overview

Mix equal volumes of A and B reagents {Luminol Reagent +Peroxidaze Reagent]. I have used up to 0.250mL each and add it to the membrane. Adjust the volume of reagents to requirement. Incubate the mixture for 2min.Cover blot with clear plastic wrap or sheet protector and expose either with xray film or CCD camera.

Tips

Use blots that are washed appropriately after secondary to avoid back ground.
