CD4+ T cells were extracted from the spleens and lymph nodes of mice, selected for CD4 using a negative selection column, and then incubated for 5 days with cytokines and antibodies to skew the cells towards a TH17 phenotype before being tested for IL-17 production via flow cytometric analysis.
