Damage free excision of DNA bands required for sensitive cloning applications (wherein no mutations in downstream constructs are required) from 1.2 % agarose gels was excised in sterile manner using FastGene Blue Light LED Trans-Illuminator. Large scale DNA bands excision from agarose gel can't be done under UV illuminator as it can potentially cause mutation in DNA to be used as well hazardous to operator. In such case, Blue LED Trans-Illuminator is the best option.

Protocol Overview

Using FastGene Blue Light LED trans illuminator for excision of DNA bands from agarose gels is pretty straight forward protocol. Setting up of equipment is very simple, just need to plug the equipment to power supply. Open illuminator's top lid and then place agarose gel containing DNA (which is been run through electrophoresis apparatus). Switch on LED illuminator, which will expose gel to Blue Light. DNA bands are visualised as bright bands depending on concentration of DNA in each well. LED illuminator generates uniform blue light and it is sufficient enough to visualise DNA bands. Then excise the DNA bands and proceed with purification protocol. After excision, power off the equipment.

Results Summary

Satisfactory excision of DNA bands from gels. I regularly perform large scale DNA excision from gels which are intended for sensitive cloning applications, where I want to minimise any mutations due to exposure of UV light (which is possible if done in UV illuminator). FastGene Blue Light LED trans illuminator is the best option for such type applications.
