During the sample preparation for next-generation sequencing, libraries are ligated to the ion spheres in such a way that only one oligo is bound to one ionosphere. The oligo is amplified on the ion sphere by ion one touch PCR. After the amplification, the ion spheres have to be enriched and purified for sequencing. Dynabeads™ MyOne™ Streptavidin C1 were used to enrich the ion spheres and purify the amplified spheres.
Purification of the biotinylated nucleic acids
Ion one touch amplified ionspheres
Around 100 microliters of the Dynabeads were used for the purification of the biotinylated ion spheres. The amplified ion spheres were incubated with Dynabeads for a specific incubation time. After, the beads were washed with Tween solution and finally with the wash solution for a couple of times. Finall, the biotinylated beads were eluted using elution buffer.
The kits can be used without any problems.
A good quality of the enriched sphere was obtained which can be used for sequencing purposes.
