I use SYBR Safe DNA gel stain in 1% agarose gels to visualize E. coli and S. enterica PCR products and get very good visualization in Gel imager and as well in Blue light LED illuminator. Gel extracted PCR products were used for cloning and found no mutagenic effect been induced to SYBR Safe DNA gel stain.

Starting Material

SYBR Safe DNA gel stain in 1% agarose gel and 500 ng of DNA loaded into each well of agarose gel

Protocol Overview

Prepare 1% agarose gel using SYBR safe DNA gel stain at 1:10,000 dilution in a 1x Tris Acetate EDTA buffer. Once gel is solidified, remove the gel combs while gel still in gel tank with 1X TAE buffer. Then load your DNA samples along with DNA ladder and run your agarose gel in electrophoresis apparatus at 100 volts for 30 min. Visualize gel in BioRad Gel imager or Blue light LED illuminator.

Tips

Add SYBR Safe DNA gel stain to molten agarose at 65 degree C. Protect concentrated gel stain from light exposure.

Results Summary

Good visualization of DNA bands both in BioRad Gel imager and as well in Blue Light LED illuminator which is sufficient enough to perform gel excision and purification using gel extraction kits. Less or no mutagenic effect of SYBR Safe DNA gel stain in E. coli and S. enterica DNA samples been extracted for gels stained with this stain.
